ADP-ribosylation of actin by clostridial toxins
نویسندگان
چکیده
umber of clostridial bacteria produce toxins which ADP-ribosylate monomeric actin. These include botulinum C2 toxin (2), Clostridiumperfringens iota toxin (23, 26), Clostridium spiroforme toxin (20, 27), and an ADP-ribosyltransferase produced by Clostridium difficile (21). The toxins have turned out to be valuable tools for investigating the actin cytoskeleton since ADP-ribosylation of actin blocks its polymerization. Furthermore, ADP-ribosylated actin acts as a capping protein that binds to the fast-growing barbed end of polymerized F-actin to further inhibit polymerization of nonmodified actin. This class of newly isolated toxins can be used to investigate the dynamic turnover of Gand F-actin in living cells. The best studied example of these new toxins is botulinum C2 toxin, which is described in this review in more detail.

ADP-ribosylation of the 1:1 (G-A) and 1:2 (G-A-A) gelsolin-actin complexes by Clostridium perfringens iota toxin and Clostridium botulinum C2 toxin was studied. Iota toxin ADP-ribosylated actin in the G-A complex from human platelets as effectively as skeletal muscle actin. The Km for NAD (4 microM) was identical for both substrates. متن کاملClostridial Binary Toxins: Iota and C2 Family Portraits
There are many pathogenic Clostridium species with diverse virulence factors that include protein toxins. Some of these bacteria, such as C. botulinum, C. difficile, C. perfringens, and C. spiroforme, cause enteric problems in animals as well as humans. These often fatal diseases can partly be attributed to binary protein toxins that follow a classic AB paradigm. Within a targeted cell, all clo...
متن کاملCellular Entry of Clostridium perfringens Iota-Toxin and Clostridium botulinum C2 Toxin
Clostridium perfringens iota-toxin and Clostridium botulinum C2 toxin are composed of two non-linked proteins, one being the enzymatic component and the other being the binding/translocation component. These latter components recognize specific receptors and oligomerize in plasma membrane lipid-rafts, mediating the uptake of the enzymatic component into the cytosol. Enzymatic components induce ...
متن کاملADP-ribosylation of Drosophila indirect-flight-muscle actin and arthrin by Clostridium botulinum C2 toxin and Clostridium perfringens iota toxin.
Purified Drosophila indirect-flight-muscle actin and arthrin, an actin-ubiquitin conjugate, were ADP-ribosylated by Clostridium botulinum C2 toxin and Clostridium perfringens iota toxin. Phalloidin treatment inhibited the ADP-ribosylation of Drosophila actin and arthrin. Like actin, the ADP-ribose-arthrin linkage was sensitive towards hydroxylamine treatment, indicating arginine as the amino ac...
متن کاملDelivery of proteins into living cells by reversible membrane permeabilization with streptolysin-O.
The pore-forming toxin streptolysin O (SLO) can be used to reversibly permeabilize adherent and nonadherent cells, allowing delivery of molecules with up to 100 kDa mass to the cytosol. Using FITC-labeled albumin, 10(5)-10(6) molecules were estimated to be entrapped per cell. Repair of toxin lesions depended on Ca(2+)-calmodulin and on intact microtubules, but was not sensitive to actin disrupt...
